The increased emergence of cereal stem rust in southern and western Europe, caused by the pathogen Puccinia graminis, and the prevalence of alternate (sexual) host, Berberis species, have regained attention as the sexual host may serve as source of novel pathogen variability that may pose a threat to cereal supply. The main objective of the present study was to investigate the functional role of Berberis species in the current epidemiological situation of cereal stem rust in Europe. Surveys in 11 European countries were carried out from 2018 to 2020, where aecial infections from five barberry species were collected. Phylogenetic analysis of 121 single aecial clusters of diverse origin using the elongation factor 1-α gene indicated the presence of different special forms (aka formae speciales) of P. graminis adapted to different cereal and grass species. Inoculation studies using aecial clusters from Spain, United Kingdom, and Switzerland resulted in 533 stem rust isolates sampled from wheat, barley, rye, and oat, which confirmed the presence of multiple special forms of P. graminis. Microsatellite marker analysis of a subset of 192 sexually-derived isolates recovered on wheat, barley and rye from the three populations confirmed the generation of novel genetic diversity revealed by the detection of 135 multilocus genotypes. Discriminant analysis of principal components resulted in four genetic clusters, which grouped at both local and country level. Here, we demonstrated that a variety of Berberis species may serve as functional alternate hosts for cereal stem rust fungi and highlights the increased risks that the sexual cycle may pose to cereal production in Europe, which calls for new initiatives within rust surveillance, epidemiological research and resistance breeding.

Oat is an important natural source of ß-D-glucan. This polysaccharide of the cell wall of selected cereals is known for a number of health-promoting effects, such as reducing the level of cholesterol in the blood serum, stabilizing the level of blood glucose, or enhancing immunity. ß-D-glucan has positive effects in the plant itself. There is a lack of information available, but the storage capacity of the polysaccharide and its importance as a protective substance in the plant during mild forms of biotic and abiotic stress are described. The accumulation of ß-D-glucan during the ontogenetic development of oats (Avena sativa L.) was determined in the present work. Two naked (Valentin, Vaclav) and two hulled (Hronec, Tatran) oat varieties were used. Samples of each plant (root, stem, leaf, panicle) were collected in four stages of the plant's development (BBCH 13, 30, 55, 71). The average content of the biopolymer was 0.29 ± 0.14% in roots, 0.32 ± 0.11% in stems, 0.48 ± 0.13% in leaves and 1.28 ± 0.79% in panicles, respectively. For root and panicle, in both hulled and naked oat varieties, sampling date was the factor of variability in the content of ß-D-glucan. In stems in hulled varieties and leaves in naked varieties, neither the sampling date nor variety influenced the polysaccharide content. The content of ß-D-glucan in the leaves of hulled and naked varieties decreased during the first three stages of plant development, but in the stage of milk ripeness the amount increased. The decreasing trend during milk ripeness, was also observed in the roots of both hulled and naked oats. However, in the panicle of hulled and naked oat varieties, the content of ß-D-glucan increased during plant growth. Due to practical applications of natural resources of ß-D-glucan and isolated ß-D-glucan is useful to know the factors influencing its content as well as to ascertain the behavior of the polysaccharide during plant development.

The objective of this study was to investigate the re-emergence of a previously important crop pathogen in Europe, Puccinia graminis f.sp. tritici, causing wheat stem rust. The pathogen has been insignificant in Europe for more than 60 years, but since 2016 it has caused epidemics on both durum wheat and bread wheat in local areas in southern Europe, and additional outbreaks in Central- and West Europe. The prevalence of three distinct genotypes/races in many areas, Clade III-B (TTRTF), Clade IV-B (TKTTF) and Clade IV-F (TKKTF), suggested clonal reproduction and evolution by mutation within these. None of these genetic groups and races, which likely originated from exotic incursions, were detected in Europe prior to 2016. A fourth genetic group, Clade VIII, detected in Germany (2013), was observed in several years in Central- and East Europe. Tests of representative European wheat varieties with prevalent races revealed high level of susceptibility. In contrast, high diversity with respect to virulence and Simple Sequence Repeat (SSR) markers were detected in local populations on cereals and grasses in proximity to Berberis species in Spain and Sweden, indicating that the alternate host may return as functional component of the epidemiology of wheat stem rust in Europe. A geographically distant population from Omsk and Novosibirsk in western Siberia (Russia) also revealed high genetic diversity, but clearly different from current European populations. The presence of Sr31-virulence in multiple and highly diverse races in local populations in Spain and Siberia stress that virulence may emerge independently when large geographical areas and time spans are considered and that Sr31-virulence is not unique to Ug99. All isolates of the Spanish populations, collected from wheat, rye and grass species, were succesfully recovered on wheat, which underline the plasticity of host barriers within P. graminis. The study demonstrated successful alignment of two genotyping approaches and race phenotyping methodologies employed by different laboratories, which also allowed us to line up with previous European and international studies of wheat stem rust. Our results suggest new initiatives within disease surveillance, epidemiological research and resistance breeding to meet current and future challenges by wheat stem rust in Europe and beyond.

Adverse environmental conditions, such as various biotic and abiotic stresses, are the primary reason for decreased crop productivity. Oat, as one of the world's major crops, is an important cereal in human nutrition. The aim of this work was to analyze the effect of inoculation with two species of the genus Fusarium on the selected qualitative parameters of oat grain intended for the food industry. Artificial inoculation caused a statistically significant decrease in the content of starch, oleic, linoleic, and α-linolenic acids in oat grains compared to the control. Moreover, artificial inoculation had no statistically significant effect on the content of ß-D-glucans, total dietary fiber, total lipids, palmitic, stearic, and cis-vaccenic acids. An increase in the content of polyunsaturated fatty acids in oat grains was observed after inoculation. The most important indicator of Fusarium infection was the presence of the mycotoxin deoxynivalenol in the grain. The content of ß-D-glucans, as a possible protective barrier in the cell wall, did not have a statistically significant effect on the inoculation manifestation in the grain.

A novel high molecular weight glutenin subunit encoded by the Glu-1B locus was identified in the French genotype Bagou, which we named 1B × 6.5. This subunit differed in SDS-PAGE from well-known 1B × 6 and 1B × 7 subunits, which are also encoded at this locus. Subunit 1B × 6.5 has a theoretical molecular weight of 88,322.83 Da, which is more mobile than 1B × 6 subunit, and isoelectric point (pI) of about 8.7, which is lower than that for 1B × 6 subunit. The specific primers were designed to amplify and sequence 2476 bp of the Glu-1B locus from genotype Bagou. A high level of similarity was found between the sequence encoding 1B × 6.5 and other x-type encoding alleles of this locus.

In human nutrition, oats (Avena sativa L.) are mainly used for their dietary fiber, ß-D-glucans and protein content. The content of ß-D-glucans in oat grain is 2-7% and is influenced by genetic and/or environmental factors. High levels of this cell walls polysaccharide are observed in naked grains of cultivated oat. It the work, the relationship between the content of ß-D-glucans in oat grain and the infection with Fusarium graminearum (FG) and Fusarium culmorum (FC) was analyzed. The hypothesis was that oats with higher content of ß-D-glucans are better protected and the manifestation of artificial inoculation with Fusarium strains is weaker. In the 22 oat samples analyzed, the content of ß-D-glucans was 0.71-5.06%. In controls, the average content was 2.15% for hulled and 3.25% for naked grains of cultivated oats. After the infection, a decrease was observed in all, naked, hulled and wild oats. As an evidence of lower rate of infection, statistically significant lower percentage of pathogen DNA (0.39%) and less deoxynivalenol (DON) mycotoxin (FC infection 10.66 mg/kg and FG 4.92 mg/kg) were observed in naked grains compared to hulled where the level of pathogen DNA was 2.09% and the average DON level was 21.95 mg/kg (FC) and 5.52 mg/kg (FG).

In this study, a total of 299 grain samples of wheat were collected from four production regions: the maize, sugar beet, potato and feed sectors of Slovakia. The samples were analyzed for deoxynivalenol (DON) content by using an enzyme-linked immunosorbent assay Ridascreen® Fast DON. Analysis of variance revealed a significant difference between years in DON contents (p < 0.027). The occurrence of samples with DON was 82.2% in 2010, with maximum DON content of 7.88 mg kg⁻¹, and 70.7% in 2011, with maximum DON content of 2.12 mg·kg⁻¹. The total mean DON content was 0.62 mg·kg⁻¹; in the feed region 0.22 mg·kg⁻¹; 0.63 mg·kg⁻¹ in the maize region; 0.78 mg·kg⁻¹ in the sugar beet region; 0.45 mg·kg⁻¹ the potato region. The limit of 1.25 mg·kg⁻¹ imposed by the European Union (EU) for DON content was exceeded in 13.7% of the studied samples. The average monthly rainfall for May to June played a critical role in DON content of wheat grains for maize and sugar beet producing regions. The present results indicate that DON content was at a high level in grains from wheat grown during 2010.
